Cucumber Cucurbitacin B, the Chemotherapy Enhancer for the Treatment of Lymphoma

Lymphoma is a medical and chronic condition associated with abnormal growth of lymphocytes, a type of white blood cells of the immune system that fight infection.

Lymphocytes included natural killer cells T cells and B cells are found in the lymph nodes, spleen, thymus, bone marrow, and other parts of the body.

The condition can be classified into 2 types
* Non-Hodgkin is the common form of lymphoma found in most patients.
* Hodgkin

According to the statistics, in the US, this year, approximately, 8,000 people will be diagnosed with Hodgkin lymphoma, leading to the death of 1,000 patients with the disease.

Believe it or not, Hodgkin's lymphoma is treatable, especially in its early stages. The 5-year survival rate for all patients diagnosed with Hodgkin's lymphoma is 92%.

Most cases of non-Hodgkin lymphoma are found in adults 65 years of age and over compared to the age of 15 and 40 or older than 55 in Hodgkin lymphoma.

People with a weakened immune system, autoimmunity, viral infectious disease, family history, exposure to benzene and radiotherapy, and past history of Hodgkin or non-Hodgkin lymphoma are some factors that may increase the risk of the disease.

Some researchers suggested that genetic proposition may have a strong implication with the onset of lymphoma.

Dr. Robert E. Ferrell, the lead scientist wrote, "Hereditary or primary lymphedema is a developmental disorder of the lymphatic system which leads to a disabling and disfiguring swelling of the extremities"

And, " Hereditary lymphedema generally shows an autosomal dominant pattern of inheritance with reduced penetrance, variable expression and variable age at onset".

The cucumber plant is a species of Cucumis Sativus, belongs to the family Cucurbitaceae, and native to Western Asia. It is a creeping vine with roots in the ground and grows up with the support of frames.

On finding a natural and potential compound for the treatment of lymphoma, researchers examined the effect of Cucurbitacin B (CuB) as ATO and STAT3 inhibitor against lymphoma.

Where Arsenic trioxide (ATO) is a chemotherapy drug used for the treatment of acute myeloid leukemia.

And STAT3 is a protein that regulates genes involved in cell growth and division.

Injection of CuB showed to eliminate the apoptosis resistance of Burkitt's lymphoma Ramos cells to ATO synergistically.

In other words, CuB enhanced the drug effects in the inhibition of lymphoma, particularly in resistance to Burkitt's lymphoma Ramos cells.

The results were attributed to the bioactive compound that blocks the activation of protein STAT3 associated with cancer cell proliferation and modulating the expression of proteins involved in pro and anti-apoptosis.

Interestingly, the combination of CuB and ATO kills cancer cells without causing harm to normal cell lines.

In the nude murine lymphoma model, the synergistic effect of the combination of CuB and ATO was once again confirmed by exhibiting significant apoptosis induction and tumor growth inhibition.

Dr. Ding X, the lead scientist said, "CuB synergistically enhances the apoptosis-inducing effect of ATO by inhibiting STAT3 phosphorylation in Ramos cells".

Taken altogether, cucumber combined with primary medicine may be used for the treatment of lymphoma, pending the confirmation of the larger sample size and multicenter human study.

Intake of CuB in the form of supplement should be taken with extreme care to prevent overdose acute liver toxicity.

Cucurbitacin B Inhibits Non Small Lung Cancer Cells in Vitro and Vivo

Lung cancer is a medical condition that caused abnormal growth of the cells in the lung's tissue. Most common form of primary lung cancers is derived from epithelial cells.

Non-small lung cancer is the most common type of lung cancer that accounts for 80% to 85% of all cases of lung cancers, usually grows and spreads methodically and predictably.

Most common types of lung cancers include
* Squamous-cell carcinoma often found in an outer area of the lung

* Large-cell carcinoma found in the center of the lung by an air tube

* Adenocarcinoma found in any part of the lung.

According to the statistics provided by the American cancer society, lung cancer is a leading cause of death among both men and women. Most people diagnosed with lung cancer are 65 or older, compared to a very small number of people diagnosed are younger than 45.

Most common prevalent factors of non-small cell lung cancer include

1. Smoking
Smoking is a major cause of lung cancer by enhancing oxidation stress by combining the inhaling air with cancer-causing chemical substances (carcinogens) in the cigarette.
Normally, our body can withstand and repair this damage, but prolonged exposure your lung to these danger carcinogens can result in lung cancer development.

2. Second-hand smoke
Prolonged exposure to second-hand smoke can also increase the risk of lung cancer.

3. Exposure to certain chemicals
There is an increased risk of lung cancer for people who inhale or exposing certain industrial chemical such as radon gas, arsenic, chromium, nickel, etc. in the workplace.

4. Family history
Risk of lung cancer increased if one of the members of your direct family has lung cancer.

5. Lung disease
Risk of ling can caner increase if you have or previously had certain types of lung disease

6. Excessive drinking
According to the study by Freudenheim et al., R. Curtis Ellison MD suggested that alcohol intake of more than 2 drinks per day is associated with a small increased risk of lung cancer. This study, like others, suggests a weak, positive association between consuming larger amounts of alcohol (>2 drinks a day) and lung cancer risk.

Out of many risk factors, some researchers aging may be one of the major condition as most patients with lung cancer was diagnosed with the age of 65 and over.

Dr. Federico Venuta, the lead researcher wrote, "The respiratory system undergoes a number of structural, physiological and immunological changes with age. For this reason, at this stage of our life, it is not always easy to differentiate disease from a normal state".

And, "From the epidemiological point of view, early-stage lung cancer is observed more frequently in elderly patients; in fact, stage I disease increases from 79% in patients younger than 65 years to 87% in those above 75 years (25). Also, resectability obviously increases with age (1,26): from 15.3% in those younger than 54 years to 25% above 75 years".

The cucumber plant is a species of Cucumis Sativus, belongings to the family Cucurbitaceae and native to Western Asia. It is a creeping vine with roots in the ground and grows up with the support of frames.

With an aim to find a natural and potential compound for the treatment of non-small lung cancer (NSCLC) with no side effects, researchers investigated Cucurbitacin B (CuB), a natural compound isolated from cucumber and other vegetables anti-NSCLC in vitro.

Four NSCLC cells (H1299, A549, HCC-827, and H661) in vitro and the highly aggressive H1299 xenograft in vivo were included in the study.

According to the tested assays, CuB significantly induced G2/M cell-cycle arrest and mitochondrial apoptosis.

Injection by the thiol antioxidant N-acetylcysteine (NAC) in the experiment showed to attenuate all cellular effects of CuB completely.

Interestingly, pretreatment with glutathione synthesis inhibitor butithione-sulfoxime (BSO), significantly exacerbated CuB's cytotoxic effects against the tested cancer cells in both vitro and vivo.

Further analysis of the anti-NSCLC efficacy of CuB, researchers found that CuB induces cell death via rapidly and significantly decrease the levels of protein thiols and GSH/GSSG ratio by disrupting cellular redox balance induced cytotoxicity.

Dr. Kausar H, the lead scientist said, "CuB interacts with and modulates cellular thiols to mediate its anti-cancer effects. Collectively, our data shed new light on the working mechanisms of CuB and demonstrate its therapeutic potential against NSCLC".

Furthermore, CuB also exerted anti-NSCLC in vitro and vivo CuB by exhibiting a strong anti-migratory and anti-invasive ability against metastatic NSCLC at nanomolar concentrations.

In other words, CuB showed significant tumor angiogenesis-inhibitory effects by the inhibition of migratory, invasive, and tube-forming capacities of human umbilical vein endothelial cells.

Moreover, in experimental EMT model CuB also suppressed the overexpression of protein associated with the production of proinflammatory cytokines in the initiation of cellular change and Wnt/β-catenin signaling axis in stimulating the production of formation of new blood vessels to facilitate cancer proliferation.

Taken altogether, cucumber processed a high amount of CuB may be considered supplements for the prevention and treatment of non-small cell lung cancer, pending the confirmation of the larger sample size and multicenter human study.

Intake of CuB in the form of supplement should be taken with extreme care to prevent overdose acute liver toxicity.

Fennel, a Natural Anti Fever Functional Food

Scientists may have found food and plant processes the antipyretic activity comparable to those of acetaminophen in patients with fever, some studies suggested.

Fever is a natural reaction caused by the production of pro-inflammatory cytokines of the immune system in protecting our body from damage and injury against foreign pathogens during the acute phase of infection.

According to medical literature, a person is not considered to have a significant fever until the temperature is above 100.4 F (38.0 C).

Although fever is a common syndrome, however, if you or your kid are experiencing fever accompanied by symptoms of a severe headache, unusual skin rash, sensitivity to bright light, persistent stiff neck and pain, mental confusion, particularly persistent vomiting, and difficulty breathing or chest pain, you or your kid should go to the hospital right the way.

Fennel (Foeniculum vulgare) is a plant species of the genus Foeniculum, belonging to Apiaceae (Umbelliferae), native to the Mediterranean.

The plant and food have been used in traditional and herbal medicine as warming, carminative, antispasmodic, antidepressant agent and to stimulate the appetite, ease indigestion, soothe coughing, reduce intestinal spasms, regulate the menstrual cycle, and relieve PMS,...

In the study to evaluate the antipyretic effect of the aqueous extract of the herbal coded formulation containing an equal amount of Salix alba, Emblica officinalis, Glycyrrhiza glabra, Adhatoda vasica, Viola odorata, in a yeast induced pyrexia model in rabbits, researchers found that after recording the results immediately before the administration of the extractor paracetamol and again at 1 hour, application of the extract at 240mg/kg dose significant reduces elevated temperature as compared with that of standard drug paracetamol (150mg/kg).

Fennel extract applied alone showed antipyretic activity against hyperpyrexia in mice, induced by S.C. administration of 2 mL/100 g of a 20% aqueous suspension of brewer's yeast.

As an antipyretic agent, ethanolic extract of F. vulgare fruit showed a moderate antipyretic activity that was statistically significant after 30 and 90 min.

These results showed that herbal coded medicine at a dose of 240mg/kg and fennel extract have marked an antipyretic activity in animal models and this was strongly supported by the ethnopharmacological uses of medicinal plants.

Furthermore, in the examine the hepatotoxicity induced by acetaminophen caused by the excessive production of N-acetyl-para-benzoquinone imine (NAPQI) in the induction of symptoms such as fever due to inflammation, researchers showed that application of trans anethole, an aromatic compound isolated from fennel exerted a significant effect in the inhibited pro-inflammatory mediators such as prostaglandin (PGE2), cytokines (TNF, IL-1) and nitric oxide (NO)thus decreasing the risk of fever caused by an overreaction of the immune system.

Application of AN at doses 125 and 250mg/kg once a day for seven days prevented the changes caused by the APAP overdose.

Additionally, researchers also discovered, AN infection prevents the overexpression of proinflammatory mediators such as NO, TNF, IL-1α, MIP-1α, and MCP-1 and induced the up-regulation of anti-inflammatory cytokines (IL-4 and IL-10), thus reducing the onset of fever.

Taken all together, there is no doubt that fennel may be considered a functional food for the treatment of inflammatory induced fever.

Healthy Food Onion in the Inhibition of Angiogenesis

Angiogenesis is the process that promotes the formation of new blood vessels involving many factors including the migration, growth, and differentiation of endothelial cells.

In normal cells, angiogenesis is important for the cells to obtain nutrients and metabolites through the small blood vessels. Angiogenesis has been found to process a therapeutic effect for the treatment of ischemic heart disease, peripheral arterial disease, and wound healing.

Dr. Papetti M, the lead scientist at the Tufts University School of Medicine said, "Normal angiogenesis ensures that developing or healing tissues receive an adequate supply of nutrients".
In cancers, angiogenesis stimulated by the tumor cells has been associated with tumors proliferation and metastasis.

More precisely, the angiogenesis process a function of a double sword, on one hand, the process provides an ideal condition for the treatment of diseases and wound healing. On the other hand, it promotes invasive and overgrowth tumor.

Dr. Papetti M continued, "tumor cells induce the formation of new blood supply from the preexisting vasculature, and this affords tumor cells the ability to survive and propagate in a hostile environment"

Collectively, " both normal and tumor-induced neovascularization fulfill the essential role of satisfying the metabolic demands of tissue".

Based on the findings some researchers suggested, by the inhibiting the process of blood vessels formation of the tumor, cancer may be curable.

Dr. Dai X, the lead researchers in the study of "Suppression of murine B-cell lymphoma growth by trichosanthin through anti-angiogenesis" showed, "trichosanthin TCS, ( an immunosuppressive and anti-tumor protein) inhibits murine B-cell lymphoma growth by anti-angiogenesis was reported. First, TCS inhibit tumor growth and prolonged survival significantly in vivo, and TCS depressed the formation of new blood vessels around the tumor in a dose-dependent manner".

The onion is a plant in the genus Allium, belongings to the family Alliaceae, a close relation of garlic. It is often called the "king of vegetables" because of its pungent taste and found in a large number of recipes and preparations spanning almost the totality of the world's cultures.

Depending on the variety, an onion can be sharp, spicy, tangy, pungent, mild or sweet.

On finding a potent ingredient which processes anti-cancer activity researchers studied the antiangiogenic activity of quercetin isolated from onion, using ex vivo, in vivo and in vitro models.

According to the tested differentiation, quercetin at non-toxic concentrations significantly inhibited the microvessel sprouting associated with cancer cell proliferation.

Furthermore, the activity of cancer cells migration, invasion and tube formation of endothelial cells involved in the process of angiogenesis.was also significantly decreased by the administration of quercetin.

Quercetin treatment inhibited ex vivo angiogenesis observed by chicken egg chorioallantoic membrane assay (CAM) and matrigel plug assay.

Moreover, quercetin suppressed vascular endothelial growth factor (VEGF) which has been found to induce the formation of a blood vessel for the progression of the tumor.

Additionally, downstream pathways associated with the promotion of survival and growth in response to extracellular signals that induce angiogenesis were also inhibited by quercetin in HUVECs.

Quercetin at the concentration of 20 mg/kg/d significantly reduced the volume and the weight of solid tumors in a prostate xenograft mouse model by targeting angiogenesis.

Quercetin was also found to inhibited the cancer cell viability and induced apoptosis in prostate cancer cells.

In other words, quercetin inhibits tumor growth and angiogenesis by targeting the proteins and signaling pathways that initiate the growth of new blood vessels.

Taken altogether, onion processed a higher mount of quercetin may be considered a remedy for the prevention and treatment of cancer, pending to the confirmation of the larger sample size and multicenter human study.

Not All Onions Process Antithrombotic and Thrombolytic Activity

Antithrombotic activity is an action that reduces the formation of blood clots.

Antithrombotics are the medicines that were used therapeutically for the prevention or treatment of dangerous coagulation, particularly in the acute thrombus.

Conventionally, medications that treat blood clots can be classified into anticoagulants and antiplatelet drugs
* Anticoagulants; The main function of anticoagulation is slow down clotting by reducing fibrin formation. However, they are also used to prevent clots from forming and growing.

* Antiplatelet agents; the main function of antiplatelet agents is to prevent platelets from clumping together. However, they also exert a similar effect to those of anticoagulation.

Thrombolytic activity is an action that dissolves dangerous clots in blood vessels, improves blood flow, with an aim to prevents damage to tissues associated with heart diseases and stroke.

Thrombolysis, also known as thrombolytic therapy, is a type of conventional medicine that performs the thrombolytic activity.

The most common agents of thrombolytic therapy are Eminase (anistreplase), Retavase (reteplase), and Striptease (streptokinase, kabikinase).

There are many factors associated with the risk of blood clots including bone fractures, major surgery, use of birth control pills, and pregnancy.

Epidemiologically, obesity, cigarette smoking, cancer, chemotherapy, and personal history of abnormal blood clotting are also the prevalent factors found in patients with thrombus.

The onion is a plant in the genus Allium, belongings to the family Alliaceae, a close relation of garlic. It is often called the "king of vegetables" because of its pungent taste and is found in a large number of recipes and preparations spanning almost the totality of the world's cultures.

Depending on the variety, an onion can be sharp, spicy, tangy, pungent, mild, or sweet.

On finding a potential compound that promotes blood thinning, researchers examined the property of onion extracts' antithrombotic effect.

According to the results of the experiment* Toyohira, one of the onion varieties showed significant antithrombotic activity both in vitro and in vivo.

* Moreover, Toyohira not only exhibited thrombolytic activity but also promoted the antiplatelet effect.

* Superkitamomiji, 2935A, and K83211, other varieties of onion showed only thrombolytic activity.

* More interestingly, quercetin, the richest flavonoid in onion, showed no correlation in the antithrombotic activity.
Based on the results, researchers strongly suggested that onion A. cepa can be classified into varieties with or without antithrombotic and thrombolytic effects.

In other words, only certain varieties of onion process antithrombotic and thrombolytic activity.

Taken altogether, certain onion varieties may be as antithrombotic and thrombolytic agents, pending the confirmation of the larger sample size and multicenter human study.

Brassica Food plants in the Prevention and Treatment of Cardiac Hypertrophy

Hypertrophy is a process or condition that increases and growth of muscle cells by increasing the muscular size achieved through exercise.

Lifting weights are the most common way to increase hypertrophy.

In other words, hypertrophy is the enlargement of an organ or tissue from the increase in the size of its cells by increasing the number of contractile proteins in each muscle fiber.

There are two types of muscular hypertrophy including the growth of muscle contraction parts or myofibrillar and the increased muscle glycogen storage or sarcoplasmic.

Myofibrillar training improves strength and speed, compared to sarcoplasmic growth that sustains more energy in endurance athletic events.

Cardiac hypertrophy is a medical condition associated with the abnormal enlargement, or thickening, of the heart muscle.

The most common causes of cardiac hypertrophy are the results of hypertension or valvular disease.

The condition can also be caused by athletic hypertrophy (a condition related to exercise), valve disease, hypertrophic cardiomyopathy (HOCM), and congenital heart disease.

The most common symptoms are shortness of breath, chest pain, and palpitation, especially during exercise.

On finding a potential compound that attenuates cardiac H9c2 cell hypertrophy, researchers examined the effects of 3,3'-Diindolylmethane (DIM) is the major product of the acid-catalyzed condensation of indole-3-carbinol (I3C), a component of extracts of Brassica food plants.

The study was conducted using an in vitro model with 1 µM angiotensin II (Ang II) in cultured rat cardiac H9c2 cells, to observe the effects of DIM on cardiac hypertrophy.

According to the tested analysis, the optimal concentration of DIM for 6, 12, and 24 h. showed a significant effect on the atrial natriuretic peptide, brain natriuretic peptide, and myosin heavy chain β mRNA expression induced by Ang II in H9c2 cells.

In other words, DIM lowers the blood pressure, promotes diuresis, natriuresis, vasodilation of the systemic and pulmonary vasculature, and reduction of circulating levels of endothelin and aldosterone and cardiac muscle contraction induced by Ang II in H9c2 cells.

Furthermore, H9c2 cells treated with DIM for 0, 15, 30, and 60 min induced by Ang II also improve the function of 5'-AMP-activated protein kinase catalytic subunit alpha-1 that regulates the activities of a number of key metabolic enzymes in protecting the cardiac cells from stresses in response to hypertrophic stimuli.

Based on the findings, Brassica food plants consist of DIM that may be used for the prevention of cardiac hypertrophy, pending the confirmation of the larger sample size and multicenter human study.

Whole Food Tomato Protects the Tissues of Stomach Against Gastric Injury

Gastrointestinal injury is an injury associated with the stomach, small bowel, colon, or rectum.

In other words, any damage to the organs of the digestive system caused by blunt or penetrating trauma is considered a gastrointestinal injury.

Gastrointestinal injury caused by blunt trauma in children is uncommon, accounting for less than 1% of all gastrointestinal injuries.

Believe it or not, in children, most common causes of gastrointestinal injury are caused by a discrete point of energy transfer such as a seatbelt (19%), a handlebar (13%), or a blow from abuse (19%), or other blows and is unique to this population, according to the Children's Hospital.

Depending on the organs damage, patients may experience different symptoms.

In small intestinal injury, patients may have symptoms of pain closed to the injured bowel segment, abdominal distension, and diarrhea, ileus, diffuse abdominal pain, nausea, and vomiting.

In large intestinal injury, patients may have symptoms of persistent focal pain in a trocar site with abdominal distention, diarrhea, and leukopenia.

In gastric/stomach injury, patients may first experience symptoms of nausea, vomiting, blood in the urine, and fever, following by abdominal pain, tenderness, distension, or rigidity.

In rectal injury, patients may have symptoms of rectal itching, pain, and rectal bleeding.

Tomato is red, edible fruit, genus Solanum, belongings to family Solanaceae, native to South America. Because of its health benefits, the tomato is grown worldwide for the commercial purpose
and often in the greenhouse.

On findings a potential ingredient for the prevention and treatment of gastrointestinal injury, researchers investigated the transgenic mini-tomato and protection against alcohol-induced gastrointestinal injury.

By examining the function of the epidermal growth factor (EGF) in maintaining the physiological function of the mucosa of the digestive tract, and to promote the healing of the gastric and duodenal ulcers, researchers chemically synthesized a tomato codon preference hEGF gene by Agrobacterium-mediated transformation.

Where Epidermal growth factor (EGF) has been found to play an essential role in wound healing by stimulating epidermal and dermal regeneration.

According to the radioimmunoassay (RIA),15 days of intragastric gavage (ig) administration of the rhEGF-containing juice of the transgenic tomato could significantly protect mice against alcohol-induced ulceration.

The Ulcer index expressed as a degree of the stomach lesion, also decreased from 42.20 +/- 18.13 to 16.25 +/- 9.57.

In other words, the transgenic tomato protects the integrity of the stomach cell against the insults of excessive alcohol consumption that damage the stomach lining tissues and speeds up ulcer healing

Taken altogether, tomato with rhEGF may be considered a dietary supplements for the prevention and treatment of gastrointestinal injury, pending a large sample size and multicenter human study.
